AP Bio Help

<p>I have a couple questions that I need help on here:</p>

<p>1: Using the Restriction Map Data for pAMP, ampicillin resistance plasmid and pKAN, kanamcycin resistance plasmid, and the Restriction Enzymes provided in data sheet #2, design an experiment to produce a recombinant DNA plasmid with both Amp and Kan resistance. Finally, design an explain an experiment to trasnform E.Coli MM 294 with this new recombinant plasmid.</p>

<p>2: Explain both of the following:
a. the mechanism of action of restriction enzymes
b. the different results you would expect if a mutation occured at recognition sites for enzyme Y</p>

<p>Hmm, well i'd cut both plasmids with the same restriction enzyme then put them together. Then i'd plate ecoli (after they had time to transform the plasmids) on a petri with kanam and amp. Colonies that grow have the recombinant plasmids.</p>

<p>Maybe i'm wrong though, that unit was at the beginning of the year for me.</p>
