I am taking the most rigorous senior schedule my school has to offer next year, and I plan on getting A's in all of them. My main school choice is the University of Washington, which is becoming increasingly more competitive to get into, due to their new preference for out-of-state students (budget cuts). I know that colleges will look at the senior schedule, but do they ever look at the first semester grades to see if the student was actually able to handle their classes?

Some request first semester grades; others do not. Depends on the school. Check their website; it should say that. All request a final high school transcript for where you enroll.
