Hi, I'm a freshman at a state school and thinking of transferring to another school next fall. I assume the GPA they want is from the first semester I was here. Do the classes I'm taking spring semester matter at all when I'm applying in regards to my final GPA?

Yes. You can read more detail on the Transfer Admissions 101 sticky thread. Some schools will ask for interim grade report from your spring semester; some will make your admission conditional on "satisfactory" completion of the spring term; some might defer your decision until they see spring grades (this latter is not very common).
