If I wait until say, March, to "Respond Now" is there any detriment?

Is "Respond Now" a requirement to save a spot even if I decide to go to a different school after I've received all my acceptances?

Once you've been accepted into UM and decide you will attend, then you do the "respond now" option. I'm waiting until April to respond.

Does housing priority change depending on when you submit the enrollment deposit?

No. It is based on a lottery. For last year, the higher number you get, the better dorm you get. You shouldn't worry too much over housing. If you don't like the dorm you got, there are a lot of upperclassmans waiting to give you their contracts. Or you can swap. I did that, and I went from Markley to West Quad.
