<p>Yes, they would “bother” looking at first semester grades, if your school does two grading periods. It would appear on your transcript.</p>

<p>It’s definitely gonna hurt your admission chances, and by a pretty big deal. If it’s senior year and this happens, you might probably even get rescinded - rejection after being accepted due to low grades.</p>

<p>In my opinion, it does look bad, but the chances of it affecting your admissions depends on other criteria. Is this your only low grade? If all of your grades are good, I see no reason to think that it would seriously hinder you at mid-ranked schools. For schools a little higher caliber, probably. </p>
