Hi! I am a Ghanaian applying to Stanford regular and will be sending 2 art supplements in both Music (Vocal: Soprano) and Visual Art. I am an A student taking the IB Diploma Course and my SAT scores are pretty decent. To what extent will sending 2 extremely good arts supplements boost my chances of of getting in.

it depends, i did my music supplement, but like if the music department is impressed (not the admissions counsel), it will look really good.
If its kind of mediocre, then they will disregard it and if it sucks, then it might work against you

Is the arts instructor's letter recommendation mandatory for the
arts supplement or can I send in a CD and arts supplement form from common app
without this letter?

i'm pretty sure that the letter is mandatory.. they say that they won't accept any incomplete supplements.

you have to send in the letter, it is part of the package.

Stanford has an arts initiative, so submitting strong arts supplements can definitely help you.
