Letter of Continued Interest and other supplementary materials

For schools which I applied via regular decision (decisions come out around late march/early april), would submitting another LOR from an employer a) even get to the schools and be reviewed on time and b) be beneficial considering my other letters are from core subject teachers?

Additionally, when should letters of continued interest be submitted for students who were deferred to RD in mid to late December?

No on the letter - you submitted your necessary letters with your app.

If you were deferred, you should already have filled out the form - asking to stay in. If you want to send a letter too, that’s fine - but you don’t need to if you checked the box to stay in the game. That’s all my daughter did last year and she was accepted (Miami).

Of course, anything the school has asked for - first semester grades - get in ASAP.

No reason to update ECs or anything else unless you did something that totally changes the game for you.
