Question about responding to quotes

When the prompt is to respond to a quote, should you directly respond to the quote in the essay?

For example, I wrote my essay for Amherst on the following quote: ''Difficulty need not foreshadow despair or defeat. Rather achievement can be all the more satisfying because of obstacles surmounted.''

Would somewhere in my essay, I say something like,
Difficulty thus does not "foreshadow despair or defeat," instead, blahblahblah this is why not.

Or should it just be implicitly worked in there?

Either way works.

keeping blahblahblah is key
