Am is supposed to focus mainly on the college of arts and science (the college i'm applying to) or can i make the focus of my essay something else like diversity or research? I mentioned my major and how research and diversity will help me as i learn at UPenn, but should i change the focus of my essay to specific classes and professors at the college of arts and science?

It’s okay to talk about research and diversity, as long as you’re talking about specific research opportunities or specific groups and organizations at the school that foster diversity, for example. Basically, you can talk about anything you want, as long as it relates back to something specific to UPenn.
