I had my interview for M&T around a month ago. It’s definitely not anything like a job interview or a STEM job interview, and I didn’t think it was rigorous at all. There are not any set questions, and the interviewer has the choice of what to ask. I thought the questions were more of a conversation starter than an actual Q&A. He started by asking the common “Tell me about yourself”, and then we started a conversation from there.
Regarding preparation, I would be careful not to overprepare, as it can make your responses seem robotic. Just treat it as if you were conversing with a respected elder. I would prepare for the “tell me about yourself” question and the “Why Penn/M&T?” question, and make sure that you have an answer if asked about your future plans after college (you don’t have to have a set plan, just need an answer for the question if asked). I would also prepare a list of questions to ask the interviewer, making sure that they aren’t common or googleable, and preferably with some regarding M&T specifically.
I believe all of the M&T interviews are done by M&T alumni, and many are very accomplished. I had a great time with my interview and good luck to your son on his! Don’t forget to thank the interviewer for their time via email after you’re done.

It is scheduled as virtual meet. Is the applicant expected to send his resume to the interviewer or is it just an informal conversation? If resume needs to be sent, how soon he should send it.

He can send his resume to the interviewer via email before the interview, as the interviewer does not get any info about the student.
I have a LinkedIn page that my interviewer already looked me up on, so I didn’t send a resume. Otherwise, I think it could help and definitely won’t hurt.
Just curious, how long was your M&T interview?
My son’s interview lasted for about 20min. It was virtual.
