The woman who gave me a call today concerning a volunteer-intern position told me that she was going to set up an interview, then said, "Remember, even if this is volunteering, it is a professional setting."

Could this mean to dress up for the interview?

Yes, and don’t be late, while you are in the professional setting don’t wear earbuds or headset, and don’t have private phone conversations where others can hear them.

I would wear the shirt/tie with dress pants and some nice shoes. I work at a free clinic, but I still wear a nice polo, crisp slacks, and comfortable shoes (on feet most of the time). IMHO, it’s always better to overdress (you can always ditch the tie) han wish that you wore something better! Best wishes! :)
